Crassostrea gigas mortality in France: the usual suspect, a herpes virus, may not be the killer in this polymicrobial opportunistic disease ArchiMer

Successive disease outbreaks in oyster (Crassostrea gigas) beds in France have resulted in dramatic losses in production, and subsequent decline in the oyster-farming industry. Deaths of juvenile oysters have been associated with the presence of a herpes virus (OsHV-1 µvar) and bacterial populations of the genus Vibrio. Although the pathogenicity of OsHV-1 µvar, as well as several strains of Vibrio has been demonstrated by experimental infections, our understanding of the complexity of infections occurring in the natural environment remains limited. Detection and description of a particular Ostreid herpesvirus 1 genotype associated with massive mortality outbreaks of Pacific oysters, Crassostrea gigas, in France in 2008 ArchiMer

Ostreid herpesvirus 1 (OsHV-1) infections have been reported around the world and are associated with high mortalities of the Pacific oyster (Crassostrea gigas). In the summer 2008, abnormal mortality rates ranging from 80% to 100% were reported in France and affected only Pacific oysters. Analyses of oyster samples collected during mortality outbreaks demonstrated a significant detection of OsHV-1 (75% of analysed batches), which appeared stronger than previous years. DNA sequencing based on C and IA regions was carried out on 28 batches of OsHV-1 infected Pacific oysters collected in 2008. Development of a polymerase chain reaction for the detection of abalone herpesvirus infection based on the DNA polymerase gene ArchiMer

A 5781-base pair (bp) fragment of genomic DNA from the Taiwanese abalone herpesvirus was obtained and showed 99% (5767/5779) homology in the nucleotide sequence and 99% (1923/1926) in the amino acid sequence with the DNA polymerase gene of the abalone herpesvirus strain Victoria/AUS/2007. Homology of the amino acid sequence with the DNA polymerase of ostreid herpesvirus 1 was 30% (563/1856). In this study, a PCR-based procedure for detecting herpesvirus infection of abalone, Haliotis diversicolor supertexta, in Taiwan was developed. The NAHMS Equine '98 Study was designed to provide information about the nation's equine population. Twenty-eight states were included in the study. Since 2008, the French oyster-farming sector has been confronted with exceptionally high rates of mortality affecting spats (less than one year old) of the Pacific oyster, C. gigas, of between 60 and 90% at all rearing sites and in Pertuis Charentais sea. The objective of this document is to provide an overview of the research and studies carried out by Ifremer in Poitou-Charentes since the emergence of this phenomenon. The actions presented were funded by IFREMER, and CPER Poitou-Charentes 2007-2013.

P>Summer mortality is a phenomenon severely affecting the aquaculture production of the Pacific oyster (Crassostrea gigas). Although its causal factors are complex, resistance to mortality has been described as a highly heritable trait, and several pathogens including the virus Ostreid Herpes virus type 1 (OsHV-1) have been associated with this phenomenon. A QTL analysis for survival of summer mortality and OsHV-1 load, estimated using real-time PCR, was performed using five F-2 full-sib families resulting from a divergent selection experiment for resistance to summer mortality. Le réseau d’observations conchylicoles RESCO assure, depuis 2009, le suivi de lots sentinelles d'huîtres creuses Crassostrea gigas sur des sites ateliers disposés sur l'ensemble du littoral français. Leur suivi permet d'acquérir des données nationales de croissance et de mortalité, de traduire la dynamique spatio-temporelle des performances d’élevage et ainsi de participer à la compréhension des phénomènes observés. The French network RESCO was developed in 2009, following the widespread mortalities in French oysters stocks of Crassostrea gigas that killed a majority of young cultivated oysters in summer 2008. The main objective of this network was to develop a tool of estimation of the mortalities at national scale, through a standardized monitoring of sentinel oysters In 2013, similar batches of spat and 18 month old oysters from different origins were deployed in thirteen sites located all along the French coasts to measure the spatial and temporal evolution of the mortality and growth rates.
